It should be "Can you" ?

Can you speak English can be taken as an offensive type of term in the way you say it, but most people wouldn't take offense to it.

It depends on what you're really asking. I guess do would fit if you're asking if he actually can speak English, but "Can you" would sound more grammatic to me.

It depends on how you say it. It's not offensive if you ask politely not shout it like CAN YOU SPEAK ENGLISH?!?!

Can describes the ability, while do describes the preferances of a person. Both are correct.
